Storage: The Giemsa dye solution has a shelf life of at least three years when stored at room temperature. The working solution is ready to use and should be stored at 2–8°C for up to one month.
Introduction to Giemsa Working Solution:
Giemsa staining solution is composed of two main components: Azure and Eosin. These dyes interact differently with various cellular structures based on their chemical properties. For example, eosinophilic granules, which are basic in nature, bind with the acidic dye Eosin and appear pink. On the other hand, basophilic components such as nucleoproteins or lymphocyte cytoplasm, being acidic, combine with the basic dye Azure and stain purple or blue-violet. Neutral particles, which are in an isoelectric state, can bind with both Eosin and Methylene Blue, resulting in a lavender color.
The distinct staining patterns allow for clear differentiation of cellular components under a light microscope. The Giemsa staining solution produced by Yuanmu Bio is made using imported Giemsa dye, ensuring that nuclei are stained purple or blue-violet, while the cytoplasm appears pink. This results in clear and sharp cell images, making it ideal for examining blood smears and distinguishing between different types of blood cells. It offers excellent staining performance, strong staining power, and vibrant coloration.
Staining Procedure (for reference only):
To prepare the working solution, mix 1 ml of Giemsa Concentrate with 9 ml of Giemsa Diluent and stir well before use.
1. Prepare a blood smear using standard techniques. Once the smear is dry, fix it in methanol for 2–3 minutes. Ensure the smear is of appropriate thickness and evenly distributed to avoid uneven staining.
2. Place the blood or bone marrow smear on a staining rack. Add the diluted staining solution to cover the entire smear and incubate at room temperature for 15–30 minutes.
3. Gently rinse the slide with tap water from one end, avoiding direct rinsing of the smear. Allow the slide to air dry before observing under a microscope.
Important Notes:
1. For safety, always wear a lab coat and disposable gloves when handling the staining solution.
2. If the staining is too dark or too light, adjust the staining time or the concentration of the working solution accordingly.
3. The stock solution should be prepared using conventional methods and filtered through filter paper. Avoid any contact with water during use, as this may reduce its effectiveness.
4. pH levels significantly affect the staining outcome. Ensure all slides are clean and free from acid or alkali contamination to achieve accurate and consistent results.
